The gene encoding p16 ~ (Znk4) was originally isolated as a cyclin-dependent kinase 4 inhibitor and has recently been mapped to 9p21. The deletion or mutation of the Znk4 gene was found in many tumor cell lines. Cyclin and their synergistic catalytic protein CDK4 regulate the cell cycle of eukaryotic cells. The development of G_1 phase requires cyclin D 1 -CDK4 kinase activity. In contrast to p21 ~ (waf1), p21 ~ (waf1) inhibited all cyclin / CDK complexes in vitro whereas p16 ~ (Znk4) selectively inhibited in vitro CDK4 kinase activity.
