研究了3 种环糊精对奥美沙坦酯药物的增溶效果.建立了高效液相色谱法测定环糊精溶液中奥美沙坦酯药物含量的方法,利用相溶解度法测定了奥美沙坦酯在不同浓度的β-环糊精(β-CD) 、甲基-β-环糊精(M-β-CD) 、羟丙基-β-环糊精(HP-β-CD) 溶液中的溶解度,并绘制了相溶解度曲线.在0 ~ 15 mmol /L 环糊精浓度范围内,奥美沙坦酯的溶解度随3 种环糊精浓度的增加而线性增加,相溶解度曲线呈AL型,奥美沙坦酯与3 种环糊精均是1: 1 的比例包合; 3 种环糊精对奥美沙坦酯均有显著的增溶作用且羟丙基-β-环糊精的增溶效果最佳,其次为甲基-β-环糊精和β-环糊精.“,”The solubilization effects of β-cyclodextrins on olmesartan medoxomil(OM) was investigated in aqueous solution. A method based on high performance liquid chromatography (HPLC ) to determine OM was established in β-cyclodextrins solutions. The solubilization effect of β-cyclodextrins including β-cyclodextrin (β-CD),Methyl-β-cyclodextrin (M-β-CD) and Hydroxyl-β-cyclodextrin (HP-β-CD) on olmesartan medoxomil was assessed by phase solubility method and the phase-solubility diagrams were obtained. The solubility of OM increased linearly as a function of β-CD,M-β-CD and HP-β-CD concentrations,in the range of 0 ~ 15 mmol /L. Phase-solubility diagrams were classified as typical AL type,indicating a 1: 1 molecular complex formation for the system of β-CDs and OM. And HP-β-CD provided the best solubility enhancement,followed by DM-β-CD and β-CD.
