目的探讨激素敏感性脂肪酶(HSL)基因启动子-60C>G多态性与2型糖尿病(T2DM)高脂血症患者血浆脂质水平的相关性。方法采用限制性片段长度多态性聚合酶链反应(PCR-RFLP)和DNA测序法对94例健康对照者、77例T2DM高脂血症患者的HSL基因启动子-60C>G多态性进行分析,并检测空腹血糖和血脂水平。结果试验组空腹血糖和TC、TG、LDL-C水平均显著高于健康对照组,HDL-C水平明显低于健康对照组(P<0.05);两组均未发现HSL基因-60GG纯合型,试验组CG杂合型频率较健康对照组高,G等位基因频率也较健康对照组高,但两组间的基因型频率和等位基因频率差异均无统计学意义(P>0.05);试验组CC型血浆TC、TG、LDL-C水平均明显高于健康对照组CC型、CG型(P<0.05),HDL-C水平明显低于健康对照组CC型(P<0.05);试验组和健康对照组组内CC型和CG型血脂水平差异均无统计学意义(P>0.05)。结论 HSL基因启动子-60C>G多态性与湖北地区汉族人群中T2DM高脂血症的血脂水平无明显关联。
Objective To investigate the relationship between -60C> G polymorphism of hormone-sensitive lipase (HSL) gene promoter and plasma lipid levels in patients with type 2 diabetes mellitus (T2DM). Methods Polymorphisms of HSL gene promoter -60C> G in 94 healthy controls and 77 T2DM hyperlipidemia patients were determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and DNA sequencing Analysis, and detection of fasting blood glucose and blood lipid levels. Results The levels of fasting blood glucose, TC, TG and LDL-C in the experimental group were significantly higher than those in the healthy control group, and the levels of HDL-C in the experimental group were significantly lower than those in the healthy control group (P <0.05). No HSL-60GG homozygous , The CG heterozygous frequency in the experimental group was higher than that in the healthy control group, and the G allele frequency was also higher than that in the healthy control group. However, there was no significant difference in genotype frequency and allele frequency between the two groups (P> 0.05) (P <0.05). The levels of TC, TG and LDL-C in model group were significantly higher than those in healthy control group (P <0.05). The levels of HDL-C in CC group were significantly lower than those in healthy control group There were no significant differences in CC and CG blood lipid levels between the experimental group and the healthy control group (P> 0.05). Conclusion There is no significant correlation between -60C> G polymorphism of HSL gene promoter and blood lipid level of T2DM hyperlipidemia in Han nationality in Hubei Province.
