【目的】观察丹红注射液对动脉粥样硬化(AS)模型家兔内皮功能及肝X受体(LXR)mRNA表达的影响。【方法】将60只新西兰雄性白兔随机均分为正常对照组、模型组、丹红注射液组;除正常对照组外,模型组和丹红注射液组家兔均采用喂饲高胆固醇饲料联合注射牛血清白蛋白的方法复制AS模型,丹红注射液组给予肌注丹红注射液(0.3 m L·kg~(-1)·d~(-1))。在第1周和第8周取材,采血并分离肝脏和左髂外动脉。运用酶法检测各组家兔血浆一氧化氮(NO)、血管性血友病因子(v WF)、内皮素-1(ET-1);实时荧光定量聚合酶链反应法检测各组家兔肝脏肝X受体α(LXRα)mRNA和肝X受体β(LXRβ)mRNA;组织形态学分析各组家兔左侧髂外动脉AS斑块形成情况。【结果】第1周时,3组家兔血浆NO、v WF、ET-1水平及LXR mRNA表达比较,差异无统计学意义(P>0.05)。第8周时,与正常对照组比较,模型组血浆NO水平显著降低(P<0.01),v WF、ET-1水平显著升高(P<0.01),LXRαmRNA表达显著升高(P<0.01),LXRβmRNA表达差异无统计学意义(P>0.05),血管AS病变明显;与模型组比较,丹红注射液组血浆NO水平显著升高(P<0.01),v WF、ET-1水平显著降低(P<0.01),LXRαmRNA表达显著降低(P<0.01),LXRβmRNA表达差异无统计学意义(P>0.05),血管AS病变明显减轻。【结论】丹红注射液能够改善AS家兔的血管内皮功能,延缓AS板块的进展,其机制可能与调控肝脏LXRα表达的作用有关。
【Objective】 To observe the effects of Danhong injection on endothelium function and hepatic X receptor (LXR) mRNA expression in rabbit model of atherosclerosis (AS). 【Methods】 Sixty male New Zealand white rabbits were randomly divided into normal control group, model group and Danhong injection group. Except normal control group, rabbits in model group and Danhong injection group were fed with high cholesterol diet The AS model was duplicated by injecting bovine serum albumin, and the Danhong injection group was given Danhong injection (0.3 m L · kg -1 d -1). Draw on week 1 and week 8 and collect blood and isolate the liver and left external iliac artery. Plasma nitric oxide (NO), von Willebrand factor (vWF) and endothelin-1 (ET-1) were detected by enzyme-linked immunosorbent assay (real-time fluorescent quantitative polymerase chain reaction) Liver LXRα mRNA and LXRβ mRNA were detected by immunohistochemistry. Histomorphology was used to analyze the formation of AS plaques on the left external iliac artery of rabbits in each group. 【Results】 There was no significant difference in plasma NO, vWF, ET-1 levels and LXR mRNA expression in the three groups at the first week (P> 0.05). At the 8th week, compared with the normal control group, the levels of plasma NO in the model group were significantly decreased (P <0.01), the levels of vWF and ET-1 were significantly increased (P <0.01), the expression of LXRα mRNA was significantly increased (P <0.01) (P> 0.05), the pathological changes of vascular AS were obvious. Compared with the model group, the plasma NO level in Danhong injection group was significantly increased (P <0.01), and the levels of vWF and ET-1 were significantly decreased (P <0.01). The expression of LXRαmRNA was significantly decreased (P <0.01), while the expression of LXRβmRNA was not significantly different (P> 0.05). 【Conclusion】 Danhong injection can improve the endothelial function of AS rabbits and delay the progression of AS. The mechanism may be related to the regulation of LXRα expression in the liver.
