目的探讨粒细胞巨噬细胞集落刺激因子(GM-CSF)对糖尿病创面愈合的影响。方法70只C57BL/6小鼠分为野生小鼠组(对照组,n=35)和糖尿病模型组(DM组,n=35)。腹腔麻醉后在背部中线两侧各制作0.8cm×0.8cm创面。创面动态摄像并于相应时间段取标本,观察创面组织愈合情况,同时计算创面愈合率;ELISA法测定创面GM-CSF表达。结果创面形成后第3天起,DM组小鼠创面愈合率较对照组明显下降,以创面形成后7d内变化最为明显;创面形成后第1天,2组小鼠创面GM-CSF表达均明显增高;创面形成后第1天和第3天,对照组小鼠创面GM-CSF表达显著高于DM组。结论GM-CSF的低表达则可能与创面愈合早期炎症细胞浸润减少有关。
Objective To investigate the effect of granulocyte-macrophage colony stimulating factor (GM-CSF) on diabetic wound healing. Methods 70 C57BL / 6 mice were divided into three groups: the control group (n = 35) and the diabetic model group (n = 35). After intraperitoneal anesthesia in the back of the midline on both sides of the production of 0.8cm × 0.8cm wound. Wound dynamic imaging and samples taken at the appropriate time to observe wound tissue healing, while calculating the rate of wound healing; ELISA method to determine the expression of GM-CSF on the wound. Results The wound healing rate of DM group was significantly lower than that of the control group on the third day after wound formation, and the most obvious change occurred within 7 days after wound formation. On the first day after the wound formation, the expression of GM-CSF in the two groups was significant Increased; on the 1st day and the 3rd day after wound formation, the expression of GM-CSF in the control group was significantly higher than that in the DM group. Conclusion The low expression of GM-CSF may be related to the reduction of inflammatory cell infiltration in the early stage of wound healing.
