目的:研究钙增敏剂对内毒素休克大鼠心肌钙反应性的影响。方法:利用离休乳头肌灌流技术,测定大鼠高体左室乳头肌在含0.25~5.5mmol/L的H-K液中的等长收缩张力T及张力的变化速率(±dT/dtmax)的变化。结果:内毒素休克组T及±dT/dtmax。在每个钙浓度点上的值均明显低于假休克组,Ca2+-T、Ca2+-±dT/dtmax曲线向下移位。内毒素休克大鼠乳头肌经合1±10-5mol/L的MCO-154的H-K液灌流后,每个钙浓度点上的T和±dT/dtmax均明显增加,接近假休克组,明显高于内毒素休克组,Ca2+-T、Ca2+-±dT/dtmax曲线向上移位。MCI-154的上述作用明显优于甲腈吡酮,且其作用效果不被α、β受体阻断剂及钙通道阻断剂所抑制。结论:钙增敏剂可逆转内毒素休克所致的心肌收缩蛋白对钙反应性的降低,改善内毒素休克机体的心肌收缩性能。
Objective: To study the effects of calcium sensitizers on cardiac calcium reactivity in endotoxic shock rats. Methods: The contractile tension T and the rate of change of tension (± dT / dtmax) were measured in the hippocampal papillary muscles of hippocampus containing 0.25-5.5 mmol / L H-K solution by means of reperfusive papillary perfusion. Results: T and ± dT / dtmax in endotoxic shock group. The value of Ca2 + -T and Ca2 + - ± dT / dtmax shifted downwards at each calcium concentration point significantly lower than sham group. T cells and ± dT / dtmax in each point of calcium concentration increased significantly after perfused with HK solution of 1 ± 10-5mol / L MCO-154 in endotoxic shock rats, close to the sham group In endotoxin shock group, Ca2 + -T, Ca2 + - ± dT / dtmax curve shifted upward. The above-mentioned effects of MCI-154 are obviously superior to tetracinin, and its effect is not inhibited by α, β-blockers and calcium channel blockers. CONCLUSION: Calcium sensitizer can reverse the decrease of calcium response induced by endotoxin shock and improve the myocardial contractility of endotoxin shock.
