目的建立高效液相色谱法测定复方双花藤止痒搽剂中苦参碱的含量。方法采用色谱柱为Thermo ODS Hypersil(4.6mm×250mm,5μm),以乙睛-磷酸盐缓冲液(取磷酸二氢钾1.36g,加1mL三乙胺,加水至1000mL,用磷酸调pH至5.5)(4∶96)为流动相,检测波长220nm,流速1.0mL·min-1,柱温为35℃。结果苦参碱在0.43~21.44μg内有良好的线性关系(r=0.9999),平均回收率为100.40%,RSD为1.93%(n=6)。结论该方法结果准确可靠,重复性好,可用于复方双花藤止痒搽剂中苦参碱的含量测定。
OBJECTIVE To establish a HPLC method for the determination of matrine in compound Shuang Hua Ting Antipruritic liniment. Methods The column was Thermo ODS Hypersil (4.6mm × 250mm, 5μm). The column was washed with acetonitrile-phosphate buffer (1.36g, potassium dihydrogen phosphate 1.36g, 1mL triethylamine and water to 1000mL, adjusted to pH 5.5 with phosphoric acid ) (4:96) as the mobile phase at a detection wavelength of 220 nm at a flow rate of 1.0 mL · min-1 with a column temperature of 35 ° C. Results There was a good linear relationship between matrine and 0.43 ~ 21.44μg (r = 0.9999). The average recovery was 100.40%. The RSD was 1.93% (n = 6). Conclusion The method is accurate and reliable with good reproducibility and can be used to determine the content of matrine in Compound Shuanghua Ting Antipruritic liniment.
