为了得到高转移率的小鼠肺癌体外传代细胞株,将Lewis肺癌实体瘤组织在体外进行14个月的传代培养,获得了L3-8细胞株。对该细胞生物学特征的研究结果表明,第39代和第65代细胞的倍增时间分别为23.5h和26.2h;第80代细胞的平皿和软琼脂集落形成率分别为30.7%和21.2%;在C57BL小鼠体内的成瘤率为100.0%,肺转移率为83.3%,平均转移灶2.9个。此细胞株较原体内传代株具有更高的转移率,为肿瘤细胞生物学、免疫学和药理学等的研究提供了一种良好的实验模型
In order to obtain a high transfer rate mouse lung cancer in vitro passage cell line, solid tumor tissue of Lewis lung cancer was subcultured in vitro for 14 months to obtain L3-8 cell line. The results of the biological characteristics of the cell showed that the doubling times of the 39th and 65th generation cells were 23.5 h and 26.2 h, respectively; the plateau and soft agar colony formation rates of the 80th generation cells were 30.7%, respectively. And 21.2%; in the C57BL mice, the tumor formation rate was 100.0%, the lung metastasis rate was 83.3%, and the average metastasis was 2.9. This cell line has a higher transfer rate than the original strains in vivo, providing a good experimental model for the study of tumor cell biology, immunology and pharmacology.
