目的探讨锌对生长期大鼠海马Uch-L1表达水平的调控作用。方法雄性断乳Wistar大鼠36只,按体重随机分为对照组(ZA)、缺锌组(ZD)和缺锌对喂组(PF),ZD组喂饲含锌量1.5mg/kg的缺锌饲料,PF组和ZA组喂饲含锌量30mg/kg的足锌饲料,PF组摄食量与ZD组一致。喂饲4周后,测定血浆锌含量和碱性磷酸酶活性以评价机体锌状况,用避暗试验测定各组大鼠的学习记忆能力,采用Western blot和RT-PCR方法检测各组大鼠海马Uch-L1蛋白及其mRNA的表达水平。结果ZD组血浆锌含量及碱性磷酸酶活性均显著低于PF组和ZA组;ZD组大鼠在避暗试验中的进洞潜伏期显著低于PF组和ZA组;与PF组和ZA组相比,ZD组大鼠海马Uch-L1及其mRNA表达水平明显降低。结论缺锌可下调大鼠海马Uch-L1表达水平。
Objective To investigate the regulatory effect of zinc on the expression of Uch-L1 in the hippocampus of rats during the growth phase. Methods Thirty - six male Wistar rats were randomly divided into control group (ZA), zinc deficiency group (ZD) and zinc deficiency to feed group (PF). ZD group was fed with 1.5 mg / kg zinc deficiency Zinc diet, PF group and ZA group fed enough zinc diet containing 30mg / kg of zinc, PF group intake and ZD group. After 4 weeks of feeding, the levels of zinc and alkaline phosphatase in plasma were measured to evaluate the zinc status of the rats. The learning and memory abilities of rats in each group were determined by dark-avoidance test. The hippocampus Uch-L1 protein and its mRNA expression levels. Results The levels of zinc and alkaline phosphatase in ZD group were significantly lower than those in PF group and ZA group. The cave penetration latency in ZD group was significantly lower than that in PF group and ZA group Compared with control group, the expression of Uch-L1 and its mRNA in hippocampus of ZD group decreased significantly. Conclusion Zinc deficiency can down-regulate the expression of Uch-L1 in hippocampus of rats.
