目的 对所建立的人皮肤成纤维细胞系进行安全性检测。方法 对人皮肤成纤维细胞进行形态学观察、染色体核型分析、软琼脂试验、裸鼠致瘤试验、内毒素试验、支原体检测、病毒因子检测、细菌、真菌检测、异常毒性试验。结果 结果均为阴性。结论 所获取的皮肤成纤维细胞可作为一种安全、可靠的靶细胞用于基因治疗研究。
Objective To detect the safety of human dermal fibroblast cell line established. Methods Human dermal fibroblasts were observed morphologically, karyotype analysis, soft agar assay, tumorigenicity test in nude mice, endotoxin test, mycoplasma test, viral factor test, bacteria and fungi test and abnormal toxicity test. The results were negative. Conclusion The obtained dermal fibroblasts can be used as a safe and reliable target cell for gene therapy research.
