本研究通过对大鼠结肠粘膜组织学观察,过氧化脂质(LPO)、髓过氧化物酶(MPO),谷胱甘肽过氧化物酶(GSH—Px)及其线粒体ATPase水平的检测,旨在探讨表皮生长因子(EGF)和谷氨酰胺(GLN)防治大鼠乙酸性结肠炎的机理.结果表明:(l)EGF组、GLN组及EGF+GLN组结肠粘膜损伤指数均明显低于乙酸组(P<0.01);光镜、透射及扫描电镜观,EGF+GLN组结肠粘膜炎症明显轻于其余3组。(2)EGF组、GLN组和EGF+GLN组LPO、MPO水平分别明显低于乙酸组;4组间GSH—Px活性差别无显著性(P>0.05);GLN组Na~+—K~+—ATPase、Ca~(2+)—ATPase活性均显著高于乙酸组,其Ca~(2+)—ATPase活性还明显高于EGF组;EGF+GLN组Na~+—K~+—ATPase、Ca~(2+)—ATPase和Mg~(2+)—ATPase水平均显著高于乙酸组,该组后两种ATPase还明显高于EGF组。提示EGF与GLN联合应用,可增加组织营养、能量供给,抑制脂质过氧化损伤,防治炎症,从而对结肠粘膜具有明显的保护作用。
In this study, colonic mucosa histological observation of rats, detection of peroxidative lipid (LPO), myeloperoxidase (MPO), glutathione peroxidase (GSH-Px) and mitochondrial ATPase level, The aim of this study was to investigate the mechanism of epidermal growth factor (EGF) and glutamine (GLN) on the prevention and treatment of acetic acid-induced colitis in rats.Results: (1) The index of colon mucosal damage in EGF group, GLN group and EGF + GLN group were significantly lower than Acetic acid group (P <0.01); light microscope, transmission and scanning electron microscope, EGF + GLN group colitis mucositis was significantly lighter than the other three groups. (2) The levels of LPO and MPO in EGF group, GLN group and EGF + GLN group were significantly lower than those in acetic acid group. There was no significant difference in GSH-Px activity between the four groups (P> 0.05) The activities of ATPase, Ca ~ (2 +) - ATPase were significantly higher than those in the acetic acid group, and the activity of Ca ~ (2 +) - ATPase was also significantly higher than that in the EGF group; Na ~ + -K ~ + -ATPase, The levels of Ca ~ (2 +) - ATPase and Mg ~ (2 +) - ATPase were significantly higher than those in the acetic acid group, and the latter two ATPases were also significantly higher than those in the EGF group. Tip EGF and GLN combined application can increase tissue nutrition, energy supply, inhibition of lipid peroxidation damage, prevention and treatment of inflammation, which has obvious protective effect on colonic mucosa.
