目的探讨新生儿缺氧缺血性脑病血清中miRNA-21、缺氧诱导因子1(HIF-1α)的表达及临床意义,为临床上早期诊断及治疗缺氧缺血性脑病提供科学依据。方法选取2012年1月-2013年6月在新乡市中心医院和妇幼保健院出生的新生儿,分为:缺血缺氧组49例;正常对照组29例,共计78例。两组于生后24~48h内采血5mL。应用实时荧光定量PCR技术检测血清中miRNA-21和HIF-1α的表达水平。结果缺氧缺血组与对照组相比,缺氧缺血组血清中miRNA-21和HIF-1α均呈现高表达(P<0.05)。结论血清中miRNA-21和HIF-1α的高表达与缺氧缺血性脑病有关,并可作为早期判断脑损伤的有用的生物学指标。
Objective To investigate the expression and clinical significance of miRNA-21 and hypoxia inducible factor 1 (HIF-1α) in neonates with hypoxic-ischemic encephalopathy and provide a scientific basis for early diagnosis and treatment of hypoxic-ischemic encephalopathy. Methods From January 2012 to June 2013, newborns born in Xinxiang Central Hospital and Maternal and Child Health Hospital were divided into four groups: 49 cases of hypoxia-ischemia group and 29 cases of normal control group (78 cases in total). Blood samples were taken from 5 to 24 hours after birth in both groups. Real-time fluorescence quantitative PCR was used to detect the expression of miRNA-21 and HIF-1α in serum. Results Compared with the control group, the levels of miRNA-21 and HIF-1α in hypoxic-ischemic group were significantly increased (P <0.05). Conclusion The high expression of miRNA-21 and HIF-1α in serum correlates with hypoxic-ischemic encephalopathy and may be used as a useful biological indicator for early diagnosis of brain injury.
