目的:通过HPLC-DAD多谱功能,建立多波长同时检测青叶胆中7种有效成分含量的HPLC方法;优化检测时青叶胆中7种有效成分提取的最佳工艺。方法:采用HPLC-DAD扫描3D图谱,直接获得各环烯醚萜类、各酮口山及齐墩果酸的最适宜吸收波长。根据结果开启210 nm、240 nm和265 nm多波长采集数据同时检测7种有效成分;采用L_9(3~4)正交试验,以7种有效成分含量为指标,考察提取溶剂的单次液料比、单次提取时间、提取温度、提取次数等因素的影响,优化超声提取工艺。结果:7种成分在线性范围内线性关系良好,相关系数r均达到0.9999,平均回收率99%~101%,RSD均小于2%;最佳提取工艺单次液料比为10,每次提取30 min,提取温度为50℃,提取次数为3次。结论:HPLC-DAD多波长多通道检测青叶胆质量具有简便、高效、重复性好等特点;该提取工艺操作简单、合理、可行。
OBJECTIVE: To establish an HPLC method for the simultaneous detection of seven active components in Aloe Barbadensis Glabra by multi-wavelength by HPLC-DAD multi-spectral analysis and to optimize the extraction of the seven active constituents in Aloe vera. Methods: The optimum absorption wavelength of each iridoids, ketupousan and oleanolic acid was directly obtained by HPLC-DAD scanning of 3D spectra. According to the results, the data of 210 nm, 240 nm and 265 nm were acquired and seven active components were detected at the same time. The L9 (3-4) orthogonal test was used to determine the content of seven effective components. Ratio, single extraction time, extraction temperature, extraction times and other factors, to optimize ultrasonic extraction process. Results: The linear relationship between the seven components was good, the correlation coefficient r reached 0.9999, the average recovery was 99% ~ 101%, RSD was less than 2%; the best extraction process single liquid to liquid ratio of 10, each extraction 30 min, extraction temperature was 50 ℃, extraction times were 3 times. Conclusion: HPLC-DAD multi-wavelength multichannel detection of green leaf gallbladder quality is simple, efficient, good repeatability and other characteristics; the extraction process is simple, reasonable and feasible.
