紫菀根10.0kg,甲醇热提取3次,提取物用二氯甲烷和水萃取。除去溶剂,二氯甲烷部分(150g)硅胶柱层析,己烷-乙酸乙酯(9:1~1:1)梯度洗脱。20%乙酸乙酯洗脱部分(约10g)以氯仿-甲醇重结晶,得到无色结晶(2.0g)。 IR显示在3624cm~(-1)有羟基的吸收峰。MS中分子离子峰m/z 428定其分子式为C_(30)H_(52)O。因其在氯仿、乙酸乙酯、甲醇等溶剂中不溶解,故在10%的TFA-d1/CDCl_3中进
Astragalus root 10.0 kg was extracted with methanol three times and the extract was extracted with dichloromethane and water. The solvent was removed and the dichloromethane fraction (150 g) was purified by column chromatography on silica gel eluting with a gradient of hexane-ethyl acetate (9:1 to 1:1). The 20% ethyl acetate eluted fraction (about 10 g) was recrystallized from chloroform-methanol to give colorless crystals (2.0 g). IR shows a hydroxyl absorption peak at 3624 cm-1. The molecular ion peak m/z 428 in MS determined its molecular formula as C_(30)H_(52)O. Because it is insoluble in chloroform, ethyl acetate, methanol and other solvents, it enters in 10% of TFA-d1/CDCl_3.
