目的:本研究拟观察鸦胆子油乳注射液、华蟾素注射液和注射用黄芪多糖对结肠癌耐药细胞株的杀伤作用,为临床合理、有效使用中成药提供实验依据。方法:运用药物浓度梯度递增间歇诱导法建立人结肠癌lovo细胞耐5-氟尿嘧啶细胞株,并命名为lovo/5-Fu。MTT法检测lovo/5-Fu的耐药能力及不同浓度中成药对lovo/5-Fu细胞株的耐药逆转作用,并根据相应结果计算耐药逆转倍数(RF)。细胞迁移实验分为4组:空白对照组、黄芪多糖组、鸦胆子组和华蟾素组,检测不同中成药对lovo/5-Fu细胞株迁移能力的影响。结果:5-Fu对lovo细胞株的IC_(50)为14.5 mg/L,对lovo/5-Fu细胞株的IC_(50)为115.1 mg/L,根据上述两株细胞的IC_(50)可算出lovo/5-Fu细胞株对5-Fu的耐药指数为:7.94。浓度为17.3、45.3、83.8、112.5 mg/L鸦胆子油乳注射液的耐药逆转倍数分别为9.5、23.5、54.4、86.5倍;浓度为8.2、21.2、38.6、50.1 mg/L华蟾素注射液的耐药逆转倍数分别为11.2、25.2、56.6、90.5倍;浓度为26.3、70.9、115.2、163.4 mg/L注射用黄芪多糖的耐药逆转倍数分别为1.5、8.2、26.5、74.2倍。与空白对照组比较,黄芪多糖组、鸦胆子组和华蟾素组侵出小室的细胞数目明显减少,迁移能力明显减弱,其中以鸦胆子组和华蟾素组更为明显,差异均有统计学意义(P<0.05)。结论:鸦胆子油乳注射液、华蟾素注射液和注射用黄芪多糖这三种中成药均能不同程度的逆转lovo/5-Fu对5-Fu的耐药性,其中以鸦胆子油乳注射液和华蟾素注射液最为明显。
Objective: The purpose of this study is to observe Brucea javanica oil emulsion injection, cinobufacini injection and astragalus polysaccharide injection on colon cancer drug-resistant cell lines for clinical reasonable and effective use of proprietary Chinese medicine to provide experimental evidence. CONCLUSION: Brucea javanica oil injection, cinobufacini injection and astragalus polysaccharide injection all can reverse the resistance of lovo / 5-Fu to 5-Fu to varying degrees.
