目的观察牙齿移动过程中破牙骨质细胞和破骨细胞分化因子(ODF)表达的变化,研究伊班膦酸钠对大鼠正畸源性根吸收的作用。方法选取48只SPF级雌性Wistar大鼠,建立正畸牙齿移动动物模型,并设置自身对照。所有大鼠均于安装矫治器前3天在实验侧牙齿近中腭侧黏骨膜下局部注射50μL伊班膦酸钠,对照侧注射50μL生理盐水。每3天注射一次至实验结束。实验第3、7、14天各处死16只大鼠,随机选择8只作组织学切片观察,另外8只作扫描电镜(SEM)观察。结果①实验第7天和14天实验侧破牙骨质细胞数量小于对照侧(P<0.05);②实验第7天和第14天实验侧牙周组织压力侧ODF阳性表达程度低于对照侧(P<0.05);③SEM结果显示,实验侧第3、7和14天牙根表面吸收陷窝较对照侧数量少、面积小。结论局部应用伊班膦酸钠可以减少牙周组织压力侧破牙骨质细胞的数量,降低牙周组织压力侧ODF阳性表达程度,减少正畸源性根吸收的发生。
Objective To observe the changes of odontoblast and osteoclast differentiation factor (ODF) during tooth movement and to investigate the effect of ibandronate on orthodontic root resorption in rats. Methods Forty eight SPF female Wistar rats were selected to establish an animal model of orthodontic tooth movement and to set up their own control. All rats were locally injected with 50 μL of ibandronate sodium near the mid-palatal mucosa 3 days prior to the appliance installation, and 50 μL normal saline was injected into the control side. Inject every 3 days until the end of the experiment. Sixteen rats were sacrificed on the 3rd, 7th and 14th day of the experiment. Eight rabbits were randomly selected for histological examination and the other eight were observed by scanning electron microscopy (SEM). Results ① The number of unclaimed osteoblasts on the 7th day and the 14th day of experiment was smaller than that on the control side (P <0.05). ② The positive expression of ODF on the pressure side of the periodontal tissue on the 7th and 14th day was lower than that on the control side (P <0.05). ③SEM results showed that the number of resorption lacunae on the 3rd, 7th and 14th day on the experimental side was smaller than that on the control side, and the area was small. CONCLUSION: Topical application of ibandronate sodium can reduce the number of broken-line osteoclasts in the periodontal tissues, decrease the expression of ODF in the pressure side of periodontal tissues and reduce the incidence of orthodontic root resorption.
