[目的]研究osteoglycin表达与肿瘤转移的关系。[方法]构建osteoglycin反义表达载体,将其转染至小鼠肝癌Hca-P细胞,评价osteoglycin表达对Hca-P细胞转移生物学行为的影响。[结果]转染osteoglycin反义表达载体的Hca-P细胞迁移能力和侵袭能力显著提高;体内试验显示,其外周淋巴结转移率明显提高;然而细胞增殖能力及黏连能力未见明显变化。[结论]低表达osteoglycin提高Hca-P淋巴结转移能力。
[Objective] To study the relationship between osteoglycin expression and tumor metastasis. [Method] The osteoglycin antisense expression vector was constructed and transfected into mouse hepatoma Hca-P cells to evaluate the effect of osteoglycin expression on the biological behavior of Hca-P metastasis. [Results] The migration and invasion ability of Hca-P cells transfected with osteoglycin antisense expression vector was significantly increased. In vivo experiments showed that the peripheral lymph node metastasis rate was significantly increased. However, no significant changes were observed in cell proliferation and adhesion ability. [Conclusion] Low expression of osteoglycin enhances the ability of Hca-P lymph node metastasis.
