目的应用自制组织芯片检测乳癌组织中Survivin蛋白的表达。方法取21例乳腺良性增生及61例乳腺导管癌组织共82例,手工制作82点阵组织芯片,然后行Survivin免疫组化染色。以18例正常乳腺组织作对照。结果手工自行研制的组织芯片经HE及免疫组化染色后位点成功率达100.00%。正常乳腺组织中Survivin蛋白不表达,乳腺良性增生及乳腺癌组织中Survivin蛋白的阳性表达率分别为23.8%(5/21)和75.4%(46/61),3者间比较,差异有统计学意义(χ2=17.690,P<0.01)。结论在乳腺导管癌发生过程中Survivin蛋白的表达可能起重要作用。
Objective To detect the expression of Survivin protein in breast cancer by self-made tissue microarray. Methods Twenty-one cases of benign breast hyperplasia and 61 cases of breast ductal carcinoma tissues were obtained. Twenty-two dot matrix microarrays were made by hand, and then Survivin immunohistochemical staining was performed. 18 cases of normal breast tissue as a control. Results The hand-developed tissue microarray successfully demonstrated 100% success rate of HE and immunohistochemical staining. Survivin protein was not expressed in normal breast tissues, and the positive expression rate of Survivin protein in breast hyperplasia and breast cancer tissues were 23.8% (5/21) and 75.4% (46/61) respectively. The differences among the three groups were statistically significant Significance (χ2 = 17.690, P <0.01). Conclusion The expression of Survivin protein may play an important role in the carcinogenesis of breast ductal carcinoma.
