取乳猪肝细胞提取物(HE),用高效液相色谱分离纯化,观察HE及各组份对肝癌细胞BEL-7402和原代肝细胞的影响。结果显示,当 HE浓度大于 100mg/L时,对 BEL-7402细胞的 DNA合成和脱氢酶活力均有明显的抑制作用,IC_(50)(半数抑制浓度)分别为606mg/L和272mg/L。在各组分中,组分3和4的抑制作用最强,抑制活性比 HE高 10倍。50~500mg/L的 HE对正常肝细胞的酶活性和 DNA合成有明显的促进作用,其中组份1、2和5的促进作用明显。当HE浓度为1000mg/L时,对正常肝细胞的DNA合成和酶活力也有明显的抑制作用。提示,HE中存在正负双向生长调节因子。采用DNA电泳和流式细胞术结果显示,HE及组分3、4诱导BEL-7402细胞生长抑制的方式可能是细胞程序性死亡。
The hepatocyte extracts (HE) from suckling pigs were separated and purified by high performance liquid chromatography. The effects of HE and various components on hepatoma cells BEL-7402 and primary hepatocytes were observed. The results showed that when the concentration of HE was greater than 100 mg/L, the DNA synthesis and dehydrogenase activities of BEL-7402 cells were significantly inhibited. The IC 50 concentration was 606 mg/L and 272 mg/L, respectively. . In each component, components 3 and 4 had the strongest inhibitory effect and the inhibitory activity was 10 times higher than that of HE. 50-500 mg/L HE had a significant effect on the enzymatic activity and DNA synthesis of normal hepatocytes, and the promotion effect of components 1, 2 and 5 was obvious. When the HE concentration was 1000 mg/L, the DNA synthesis and enzyme activity of normal liver cells were also significantly inhibited. It is suggested that there are positive and negative bidirectional growth regulators in HE. The result of DNA electrophoresis and flow cytometry showed that HE and the components 3 and 4 induced BEL-7402 cell growth inhibition may be programmed cell death.
