目的 探讨脂质体转染胞嘧啶脱氨酶 (cytosinedeaminase ,CD)基因、放射线、树突细胞(dendriticcells,DCs)三者联合治疗直肠癌的安全有效性。 方法 应用放射线促进脂质体转染CD基因 ,再利用放射的增敏作用将直肠癌细胞杀死后与DCs和T细胞共同孵育 ,通过测定3 H标记的胸腺嘧啶糖苷摄取情况 ,观察不同DCs数目对T细胞增殖的影响并用MTT法检测不同效靶比下的肿瘤细胞的杀伤率。 结果 CD与 5 氟胞嘧啶联合放射线杀死直肠癌细胞后所产生的凋亡小体和其他肿瘤抗原可有效的被DCs递呈给T细胞 ,产生细胞毒淋巴细胞 ,对肿瘤产生特异地杀伤作用。 30∶1、15∶1、1∶1效靶比的癌细胞的杀伤率分别为 98 1%、76 2 %和 37 8%。 结论 脂质体转染CD基因、放射线、DCs三者联合具有相互协同作用 ,可显著提高对癌细胞的杀伤率
Objective To investigate the safety and efficacy of liposome-transfected cytosine deaminase (CD) gene, radiation and dendritic cells (DCs) in the treatment of rectal cancer. Methods The CD gene was transfected by lipofectamine. The rectal cancer cells were killed by irradiation and incubated with DCs and T cells. The uptake of 3 H-labeled thymidine was measured and the number of DCs was observed. On T cell proliferation and the use of MTT assay of different effect target ratio of tumor cell killing rate. Results CD and 5 flucytosine combined with radiation to kill rectal cancer cells produced by the apoptotic bodies and other tumor antigens can be DCs presented to T cells, the generation of cytotoxic lymphocytes, the tumor specific killing effect . The killing rates of cancer cells with the target ratio of 30: 1, 15: 1 and 1: 1 were 98 1%, 76 2% and 37 8%, respectively. Conclusion Liposome-transfected CD gene, radiation and DCs all have synergistic effects and can significantly increase the killing rate of cancer cells
