目的 从病人血清分离汉坦病毒Ⅰ型毒株 ,为双价疫苗的配伍提供更好的候选株。方法 从陕西HFRS患者血清 ,按常规法分离汉坦病毒。用系列单克隆抗体、空斑减少中和试验、RT PCR及部分核苷酸序列分析等检定 ,确证所分离毒株的血清 (或基因 )型别。结果 从 17份HFRS患者血清中成功地分离到 1株汉坦病毒 ,定名为PS 6株。经上述方法全面鉴定确定其为汉滩病毒 (即Ⅰ型病毒 )。该毒株有较强的繁殖力 (毒力 )及较好的抗原性 ,其免疫血清对来源于不同地区的Ⅰ型病毒有较好的中和活性。结论 PS 6毒株有可能成为Ⅰ型病毒疫苗候选株 ,为双价或多价疫苗配伍。
Objective To isolate Hantavirus type Ⅰ strain from patient serum and provide a better candidate for compatibility of bivalent vaccine. Methods The hantavirus was isolated from the serum of Shaanxi HFRS patients by routine method. A series of monoclonal antibodies, plaque reduction neutralization test, RT PCR and partial nucleotide sequence analysis and other tests to confirm the type of serum (or gene) of the isolated strains. Results One Hantavirus was successfully isolated from the sera of 17 HFRS patients and named as PS 6 strain. The above method was fully identified as Hantaan virus (ie type I virus). The strain has strong fertility (virulence) and better antigenicity, the immune serum from different regions of the type I virus has good activity and neutralization. Conclusion The PS 6 strain may become a candidate strain of type I virus vaccine and be compatible with bivalent or multivalent vaccines.
