本文应用体外双层软琼脂培养技术,对末梢血象和骨髓象正常的非青紫型先天性心脏病患儿、正常成人骨髓及小儿血液系统疾病患者的骨髓,进行了单核—巨噬细胞集落(GM-CFU-c)形成能力的观察。结果21例非青紫型先心患儿,每2×10~5骨髓有核细胞可形成31~98个集落,均值为52.92±20.46,5例正常成人骨髓的GM-CFU-c数为42.5±8.43,经t检验,两者无明显差异,先心患儿可作为实验的正常对照组。作者观察了31例急性白血病,处于发作期的20例c}M—CFU-c数目均明显减少或不生长。11例完全缓解者的GM-CFU-c均在正常水平。另1例红白血病,在骨髓象确诊前5个月,多次随访,骨髓象仅示早期细胞轻度增加,但GM-CFU-c始终生长不佳,提示正常前体细胞已有增殖受抑。GM-GFU-c培养可有助于早期诊断和较早地预报白血病的复发。本组观察10例再障中大部份明显减少。在4例恶性肿瘤骨髓转移病例,GM—CFU-c数均显著低于正常值,另外5例原发性血小板减少性紫癜和2例脾功能亢进GM-CFU-c培养结果均在正常范围。实验表明,GM-CFU-c测定对造血系统疾病的诊断、治疗及预后判断具有一定的实用价值。
In this study, double-layer soft agar culture in vitro was used to detect mononuclear-macrophage colony in the bone marrow of patients with non-cyanotic congenital heart disease with normal peripheral blood and normal bone marrow, normal adult bone marrow and pediatric hematological diseases. Another case of erythroleukemia, 5 months before the diagnosis of bone marrow like multiple visits, the bone marrow showed only a slight increase in early cells, but GM-CFU-c always poor growth, suggesting normal proliferation of pre-existing cells have been suppressed . GM-GFU-c culture can help early diagnosis and early prediction of leukemia recurrence. This group observed in 10 cases of aplastic anemia most of the obvious reduction. The numbers of GM-CFU-c were significantly lower than the normal value in the 4 cases of malignant tumor bone marrow metastasis, and the other 5 cases of primary thrombocytopenic purpura and 2 cases of hypersplenism GM-CFU-c culture results were in the normal range. Experiments show that, GM-CFU-c determination of hematopoietic system disease diagnosis, treatment and prognosis have some practical value.
