目的 :探讨去甲肾上腺素 (NA)的不同浓度、不同作用时间及肾上腺素能受体对培养 T细胞功能的影响 ,以了解神经递质 NA对细胞免疫功能的调节作用及其机制。方法 :大鼠脾细胞用刀豆素 A(Con A)刺激并培养 72 h,在刺激的开始加入 10 - 1 0~ 10 - 5 mol/L NA,或在刺激的不同时间加入 NA,或者再加入 α和 β肾上腺素能受体拮抗剂 ,然后应用噻唑蓝 (MTT)比色分析法检测 T细胞的转化反应。结果 :10 - 8~ 10 - 5 m ol/LNA均可显著抑制培养 T细胞的功能 ,其中 10 - 7m ol/L NA的抑制作用最强。 NA与先被 Con A刺激 1h后的T细胞作用 72 h左右 ,或者 NA与脾细胞作用 1h后被洗去 ,T细胞功能仍受到明显抑制 ,但 NA不能抑制先被 Con A刺激 6 h后的 T细胞转化。α肾上腺素能受体拮抗剂酚妥拉明和 β肾上腺素能受体拮抗剂心得安均可完全阻断 NA抑制 T细胞功能的作用。结论 :NA具有非浓度依赖性抑制培养 T细胞功能的作用 ,此作用可能发生于 T细胞被激活前或者 T细胞被激活早期 ,并可能通过α和β肾上腺素能受体介导。
OBJECTIVE: To investigate the effects of different concentrations of norepinephrine (NA), different time of action and adrenergic receptors on the function of cultured T cells in order to understand the regulatory effect of neurotransmitter NA on cellular immune function and its mechanism. Methods: Splenocytes of rats were stimulated with Con A and cultured for 72 h. At the beginning of stimulation, 10 - 10 ~ 5 mol / L NA was added or NA was added at different times of stimulation. Addition of α and β adrenergic receptor antagonists, and then the use of MTT colorimetric assay T cell transformation reaction. Results: The inhibitory effect of 10 - 8 ~ 10 -5 mol / L NA on cultured T cells was significantly inhibited. NA and T cells stimulated by Con A for 72 h or 72 h, or NA and spleen cells were washed away after 1 h, T cell function was still significantly inhibited, but NA can not be inhibited by Con A stimulated 6 h T cell transformation. Alpha adrenergic receptor antagonists Phentolamine and beta adrenergic receptor antagonist propranolol can completely block the effect of NA on T cell function. CONCLUSION: NA has a non-concentration-dependent inhibitory effect on the function of cultured T cells. This effect may occur before T cells are activated or when T cells are activated early and may be mediated by α and β adrenergic receptors.
