目的:分析复发性和多结节肝癌细胞P53基因突变类型,判断其细胞来源。方法:应用多聚酶链式反应(PCR)结合PCR产物测序,同时检测肝癌P53蛋白表达。结果:16例复发性肝癌中,9例计14个瘤体有P53基因突变,其中6例(38%)P53突变类型不同,属多中心来源,有3例(13%)P53突变类型相同,为单中心来源;有7例(43%)未发现P53基因突变,作者认为这些病例主要为单中心来源。在4例多结节型肝癌某同一患者的多个瘤体中发现P53具有不同突变类型。结论:肝癌复发具有多中心来源,应加强患者术后综合性治疗。
OBJECTIVE: To analyze the type of P53 gene mutation in recurrent and multinodular hepatoma cells and to determine their source of cells. Methods: Polymerase chain reaction (PCR) was combined with PCR product sequencing to detect the expression of P53 protein in hepatocellular carcinoma. Results: Among 16 cases of recurrent hepatocellular carcinoma, 9 cases including 14 tumors had P53 mutations. Among them, 6 cases (38%) had different types of P53 mutations and were multi-centered. 3 cases (13%) had the same type of P53 mutations. Single-centered sources; P53 gene mutations were not found in 7 cases (43%). The authors believe that these cases are mainly single-centered sources. P53 was found to have different types of mutations in multiple tumors in 4 patients with multi-nodular hepatocellular carcinoma. Conclusion: The recurrence of liver cancer has multi-centered sources and comprehensive treatment should be strengthened after surgery.
