目的观察血管紧张素Ⅱ(AngⅡ)对体外培养E12d小鼠肾组织各期肾小体细胞增殖和凋亡的影响。方法孕12d的母鼠40只随机分为6组,体外培养对照组(0mol/L AngⅡ)和施加不同浓度AngⅡ(10-9~10-5mol/L)组。应用免疫组织化学、原位缺口末端标记(TUNEL)法、免疫荧光双标技术及激光扫描共焦显微镜,观察体外培养24、、6d后小鼠肾组织中各期肾小体增殖细胞核抗原(PCNA)和凋亡阳性细胞的表达。结果随培养浓度增高和培养时间延长,各期肾小体细胞增殖和凋亡指数均有增加,且增殖指数在AngⅡ低浓度(10-9、10-8、10-7mol/L)、短时间(培养2、4d)时变化明显;凋亡指数在高浓度(10-6、10-5mol/L)、长时间(培养6d)时变化明显。结论AngⅡ对发育期小鼠肾小体细胞有促增殖和凋亡作用,且此作用有浓度、时间依赖性。
Objective To observe the effect of Ang Ⅱ on the proliferation and apoptosis of renal cell in vitro in E12d mice. Methods Forty pregnant female rats were randomly divided into 6 groups. The control group (0 mol / L AngⅡ) and Ang Ⅱ (10-9 ~ 10-5 mol / L) group were cultured in vitro. Immunohistochemistry, TUNEL, double immunofluorescence staining and laser scanning confocal microscopy were used to observe the expression of proliferating cell nuclear antigen (PCNA) in renal tissues after 24 and 6 days ) And apoptosis-positive cells. Results With the increase of culture concentration and the prolongation of culture time, the proliferation and apoptosis index of nephron were increased in all stages, and the proliferation index was decreased in the low concentration of AngⅡ (10-9,10-8,10-7mol / L) (Cultured for 2 days and 4 days). The apoptosis index changed obviously at high concentration (10-6,10-5 mol / L) and long time (6 days). Conclusion AngⅡ can promote the proliferation and apoptosis of rat renal somatic cells in a concentration-and time-dependent manner.
