目的 :应用人结肠癌建株LoVo细胞接种裸鼠 ,鼠间移植传代 ,建立人结肠癌裸鼠移植瘤模型。方法 :观察移植瘤大体形态和组织病理学检查 ,测定其肿瘤组织与宿主血液中的癌胚抗原 (CEA)值 ,并应用免疫组化观察肿瘤细胞中CEA分布以及153 Sm标记抗CEA单抗的荷瘤裸鼠体内放射免疫显像。结果 :移植瘤的形态和功能特性与原发肿瘤基本相似 ,移植瘤的移植成功率为 10 0 % ;放免显像显示标记抗体能浓聚于肿瘤部位 ,经腹腔注射标记抗体后 48h ,肿瘤显像清晰 ,肿瘤组织 /血液放射性比值 (T/B)为 1.42。结论 :本模型的建立为今后结肠癌放射免疫显像和治疗的实验研究提供一个理想的模型
OBJECTIVE: To establish a human colon cancer xenograft model in nude mice using human colon cancer LoVo cells inoculated nude mice and passaged in vitro. Methods: The gross morphology and histopathological examination of the tumor were observed. The CEA value in the tumor tissue and host blood was measured. The CEA distribution in tumor cells and the distribution of 153 Sm-labeled anti-CEA monoclonal antibody Radioimmunoimaging of tumor-bearing nude mice in vivo. Results: The morphological and functional characteristics of the transplanted tumor were basically similar to those of the primary tumor. The successful rate of transplanted tumor was 100%. The radioimmunoassay showed that the labeled antibody could accumulate in the tumor site. After 48h injection of labeled antibody, As clear, tumor tissue / blood radioactivity ratio (T / B) was 1.42. Conclusion: The establishment of this model will provide an ideal model for the future experimental study on radioimmunoimaging and treatment of colon cancer
