采用紫外可见分光光度计测定云南4个地区茯苓皮样品的紫外光谱,通过正交实验确定最佳提取溶液和最佳提取时间为超声50 min,获得的4个地区52个样品的紫外光谱数据,用3组平均值、2点平滑和二次微分对光谱数据进行处理,用SIMCA软件进行定性识别.结果表明:用乙酸乙酯、甲醇、氢氧化钠溶剂超声50min的提取方法最佳,实验精密度RSD分别为0~0.68%,0.074%~0.35%,0~0.48%;重复性RSD分别为0.053%~0.86%,0.042%~0.96%,0.093%~0.89%;稳定性RSD分别为0.046%~0.67%,0.063%~0.78%,0.4%~0.56%.定性分析显示,不同地区茯苓皮样品间存在较大差异.
UV spectrophotometer was used to determine the UV spectra of Poria cocos samples in four areas of Yunnan. The best extraction solution was determined by orthogonal experiment and the best extraction time was 50 min. The UV spectra of 52 samples from 4 areas were obtained. The spectral data were processed by three sets of average, two-point smoothing and second-order differential, and qualitatively identified by SIMCA software.The results showed that the optimal extraction method was 50min ultrasonic with ethyl acetate, methanol and sodium hydroxide solvent, The RSDs were 0% -0.68%, 0.074% -0.35% and 0% -0.48%, respectively. The repeatability RSD were 0.053% -0.86%, 0.042% -0.96% and 0.093% -0.89%, respectively. The RSDs were 0.046% ~ 0.67%, 0.063% ~ 0.78%, 0.4% ~ 0.56% .Qualitative analysis showed that there was a big difference between the samples of Poria cocos from different areas.
