目的:系统运用生物效应法从细胞途径探索不同黄连炮制品“止消渴”的药效差异,为黄连治疗2型糖尿病的临床有效用药提供科学依据。方法:培养3T3-L1前脂肪细胞,并诱导分化为脂肪细胞,建立胰岛素抵抗模型,观察不同黄连炮制品对细胞增殖、分化的影响,对胰岛素抵抗模型细胞培养液中葡萄糖消耗量的影响。结果:不同黄连炮制品均能明显或部分降低培养液中葡萄糖的含量,提高脂肪细胞葡萄糖的利用率,改善胰岛素抵抗,在一定剂量范围内促进3T3-L1前脂肪细胞的增值,抑制前脂肪细胞的分化;且与黄连生品相比较,萸制黄连、酒蒸黄连和酒炙黄连对上述改善作用更为明显。结论:不同黄连炮制品具有改善3T3-L1脂肪细胞胰岛素抵抗,增强脂肪细胞对葡萄糖摄取和利用的能力,从体外细胞水平上表现出改善胰岛素抵抗的作用,与黄连生品相比较,萸制黄连、酒蒸黄连和酒炙黄连“止消渴”疗效更优。
OBJECTIVE: To systematically explore the differences in the pharmacodynamic effects of different preparations of Coptis chinensis “Diabetes” from the cellular pathway by means of biological effects and to provide a scientific basis for the effective clinical treatment of type 2 diabetes mellitus. Methods: 3T3-L1 preadipocytes were cultured and induced to differentiate into adipocytes. The models of insulin resistance were established. The effects of different Coptis products on the proliferation and differentiation of cells were observed, and the effects of different concentrations of Coptis chinensis Franch on the glucose consumption in the cell culture medium of insulin resistance were observed. Results: Different Coptis products could significantly or partially reduce the content of glucose in the culture medium, improve the utilization of glucose in adipocytes, improve the insulin resistance, promote the proliferation of 3T3-L1 preadipocytes in a certain dose range, inhibit the preadipocytes Of the differentiation; and compared with Coptis crude, berberine Coptis, wine Huanglian and wine Sunburn Coptis on the above improvement is more obvious. Conclusion: Different Coptis products can improve the insulin resistance of 3T3-L1 adipocytes and enhance the ability of adipocytes to uptake and utilize glucose, and show the effect of improving insulin resistance at the cellular level in vitro. Compared with Coptis chinensis, Coptis chinensis , Wine steamed Coptis and wine Sunburn “only Diabetes ” effect is better.
