目的探讨整合素链接激酶(integrin-linked kinase,ILK)在非小细胞肺癌(non-small cell lung cancer,NSCLC)的表达情况及与临床病理因素、微血管密度(microvessel density,MVD)之间的关系。方法对101例NSCLC及相应癌旁正常肺组织行抗ILK免疫组织化学(SP法)染色;对101例NSCLC行抗CD34免疫组织化学染色并进行MVD计数。结果 ILK在正常支气管上皮细胞中为阴性表达,而在NSCLC组织中出现胞质或胞质/胞核同时表达,其总的阳性率为65.35%(66/101),与正常肺组织比较差异具有显著性(P<0.001)。无淋巴结转移组中ILK阳性率[51.16%(22/43)]显著低于淋巴结转移组[75.86%(44/58),P=0.010]。Ⅰ、Ⅱ期NSCLC患者ILK阳性率[52.27%(23/44)]显著低于其在Ⅲ、Ⅳ期患者的阳性率[75.44%(43/57),P=0.015]。NSCLC中,ILK阳性表达组的MVD值显著高于ILK阴性表达组(47.02±7.39vs.37.11±8.40,t=6.109,P<0.001)。结论 NSCLC组织中ILK表达显著高于正常肺组织,并与NSLCL的淋巴结转移,TNM分期及MVD相关,提示ILK可能通过促进血管形成而促进NSCLC的进展。
Objective To investigate the expression of integrin-linked kinase (ILK) in non-small cell lung cancer (NSCLC) and its relationship with clinical pathological factors, microvessel density (MVD) . Methods 101 cases of NSCLC and corresponding adjacent normal lung tissues were stained with anti-ILK immunohistochemistry (SP method). The anti-CD34 immunohistochemical staining of 101 NSCLCs was performed and MVD was counted. Results ILK was negatively expressed in normal bronchial epithelial cells, while cytoplasm or cytoplasm / nucleus were found in NSCLC tissues. The positive rate of ILK was 65.35% (66/101), which was significantly lower than that in normal lung tissues Significance (P <0.001). The positive rate of ILK in non-lymph node metastasis group [51.16% (22/43)] was significantly lower than that in lymph node metastasis group [75.86% (44/58), P = 0.010]. The positive rate of ILK in stage Ⅰ and Ⅱ NSCLC was significantly lower than that in stage Ⅲ and Ⅳ [52.27% (23/44)] [75.44% (43/57), P = 0.015]. In NSCLC, the MVD of ILK positive expression group was significantly higher than that of ILK negative expression group (47.02 ± 7.39vs.37.11 ± 8.40, t = 6.109, P <0.001). Conclusion The expression of ILK in NSCLC tissues is significantly higher than that in normal lung tissues. It is correlated with lymph node metastasis, TNM stage and MVD in NSLCL. It suggests that ILK may promote the progression of NSCLC by promoting angiogenesis.
