肥胖症是一种严重威胁人类健康的代谢疾病,已成为全球关注的公共健康问题。激酶S6K1是哺乳动物雷帕霉素靶蛋白(mTOR)下游一个重要的效应子,调控并介导着包括肥胖症、Ⅱ型糖尿病、癌症等在内的多种代谢疾病。作为这些疾病的重要治疗靶点,激酶S6K1已成为当前研究热点。本文主要对激酶S6K1的功能、分子调控机理、所参与调控的疾病及其小分子抑制剂的研究现状进行综述,并对其发展前景进行展望。
Obesity is a metabolic disease that is a serious threat to human health and has become a public health concern of global concern. Kinase S6K1 is an important downstream effector of mammalian target of rapamycin (mTOR) and regulates and mediates various metabolic diseases including obesity, type II diabetes and cancer. As an important therapeutic target of these diseases, kinase S6K1 has become the current research hotspot. This review summarizes the functions of S6K1 kinase, the mechanism of molecular regulation, the diseases involved in regulation and the research status of its small molecule inhibitors, and prospects its future development.
