目的用单细胞凝胶电泳技术(SCGE)研究淫羊藿总黄酮(TFE)对丝裂霉素C造成的小鼠骨髓细胞DNA损伤的保护作用。方法将50只昆明种小鼠随机分为正常对照组、模型组和TFE高、中、低3个剂量组,分别灌胃给予生理盐水或药物,连续用药7d,在第6天除正常对照组(腹腔注射生理盐水)外,其余各组均腹腔注射丝裂霉素C(MMC2mg/kg),末次给药后2h,处死小鼠,取出骨髓细胞进行单细胞凝胶电泳实验,比较各组慧星细胞百分比和慧星尾巴长度观察TFE对小鼠骨髓淋巴细胞的DNA损伤效应的影响。结果 TFE可以显著拮抗MMC所致小鼠骨髓细胞的DNA损伤,表现为慧星细胞百分比的降低和慧星尾巴长度的缩短,并有显著的量效关系。结论在本试验条件下,淫羊藿总黄酮对丝裂霉素C诱发的小鼠骨髓淋巴细胞DNA损伤具有较好的保护作用。
Objective To study the protective effect of epimedium saponin (TFE) on DNA damage induced by mitomycin C in mouse bone marrow cells by single cell gel electrophoresis (SCGE). Methods Fifty Kunming mice were randomly divided into normal control group, model group and high, medium and low TFE dose groups. Rats were given normal saline or drugs by intragastric administration for 7 days. On the 6th day, except normal control group (Intraperitoneal injection of saline), the other groups were intraperitoneal injection of mitomycin C (MMC2mg / kg), 2h after the last administration, the mice were sacrificed, bone marrow cells were removed for single cell gel electrophoresis experiments were compared Hui The effect of TFE on the DNA damage of mouse bone marrow lymphocytes was observed by the percentage of astrocytes and the length of comet tail. Results TFE could significantly antagonize the DNA damage induced by MMC in mouse bone marrow cells. The results showed that the percentage of comet cells and the length of tail of comet were shortened, and there was a significant dose-response relationship. Conclusion Under the test conditions, total flavonoids of Epimedium sagittatum against mitomycin C-induced mouse bone marrow lymphocyte DNA damage has a good protective effect.
