目的研究氯通道阻断剂5-硝基-2-(3-苯丙氨基)苯甲酸[5-nitro-2-(3-phenylpropylamino)- benzoic acid,NPPB]对人喉表皮样癌细胞(Hep-2)细胞增殖及凋亡的影响,探讨氯通道在喉癌中的意义。方法以Hep-2细胞为研究对象,采用四甲基偶氮唑蓝(MTT)比色法检测不同浓度NPPB对Hep-2细胞增殖的影响;用流式细胞术检测NPPB阻断氯通道前后Hep-2细胞凋亡率及细胞周期变化。结果NPPB浓度依赖性地抑制Hep-2细胞增殖,NPPB阻断氯通道前后Hep-2细胞凋亡率及细胞周期存在显著性差异。结论氯通道与Hep-2细胞增殖及凋亡密切相关,阻断氯通道可抑制Hep-2细胞增殖、诱导Hep-2细胞凋亡。
Objective To investigate the effect of 5-nitro-2- (3-phenylpropylamino) benzoic acid (NPPB), a chloride channel blocker, on the proliferation of human laryngeal epidermoid carcinoma cells -2) cells proliferation and apoptosis, to explore the significance of chloride channel in laryngeal cancer. Methods Hep-2 cells were used as experimental materials. The effects of different concentrations of NPPB on the proliferation of Hep-2 cells were detected by MTT colorimetric assay. Flow cytometry -2 cell apoptosis rate and cell cycle changes. Results NPPB inhibited the proliferation of Hep-2 cells in a concentration-dependent manner. There was a significant difference in the apoptosis rate and cell cycle of Hep-2 cells before and after NPPB blocked the chloride channel. Conclusion Chloride channels are closely related to the proliferation and apoptosis of Hep-2 cells. Blocking the chloride channels can inhibit the proliferation of Hep-2 cells and induce the apoptosis of Hep-2 cells.
