目的:探讨护网明目散抗实验性糖尿病视网膜病变新生血管的作用。方法:雄性Wistar大鼠100只,随机分为模型组、中药治疗组、西药治疗组、中西医结合治疗组和正常空白对照组,每组20只。以链尿佐菌素(STZ)腹腔注射诱发大鼠糖尿病模型。确定造模成功后开始分组给药,每日给药1次。用药3个月后,取各组大鼠眼球制成石蜡切片,采用免疫组化法观察视网膜血管内皮细胞生长因子(VEGF)、碱性成纤维细胞生长因(bFGF)表达水平。结果:治疗45天后,中药治疗组、中西医结合治疗组视网膜VEGF、bFGF明显低于模型组、西药治疗组(P<0.05),中西医结合治疗组与中药治疗组比较有显著差异(P<0.05)。结论:护网明目散可有效降低因微循环障碍诱发视网膜产生的视网膜VEGF)、bFGF水平,抑制新生血管的形成,阻止单纯型糖尿病性视网膜病变向增殖型的发展。
OBJECTIVE: To investigate the effect of protection net Mingmu San on experimental neovascularization of diabetic retinopathy. Methods: One hundred male Wistar rats were randomly divided into model group, traditional Chinese medicine treatment group, western medicine treatment group, integrated traditional Chinese and western medicine treatment group and normal blank control group, with 20 rats in each group. Rats with diabetes mellitus were induced by intraperitoneal injection of streptozotocin (STZ). After successful modeling, group administration was started and once a day. After 3 months of treatment, the eyeballs of each group were made into paraffin sections, and the expression of vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) were observed by immunohistochemistry. RESULTS: After 45 days of treatment, the retinal VEGF and bFGF in the Chinese medicine treatment group and the integrated traditional Chinese and Western medicine treatment group were significantly lower than those in the model group and the western medicine treatment group (P<0.05). There was a significant difference between the Chinese and Western medicine treatment group and the Chinese medicine treatment group (P< 0.05). Conclusion: Huneting Mingmu Powder can effectively reduce the level of retinal VEGF) and bFGF induced by microcirculation in the retina, inhibit the formation of new blood vessels, and prevent the development of simple diabetic retinopathy to proliferative type.
