目的通过研究三仙汤对去卵巢大鼠骨密度(BMD)、骨生物力学和血清雌二醇(E2)、孕酮(P)水平等的影响,探讨三仙汤治疗骨质疏松的机制。方法68只3月龄雌性SD大鼠随机分为7组:空白组、假手术组(Sham)、模型组(OVX)、尼尔雌醇组、以及三仙汤小、中、大3个剂量组。给药3个月后测定BMD、最大载荷(E)、破坏挠度(R)、E2、P、体质量和子宫湿重。结果骨密度:OVX组0.270g/cm2,显著低于Sham组0.291g/cm2(P<0.05),三仙中剂量较OVX有上升趋势。最大载荷:三仙中剂量117kg、大剂量组114kg与OVX组104kg相比有一定的上升趋势。破坏挠度:OVX组较Sham组下降趋势明显;三仙小、中、大3个剂量组均较OVX组显著升高(P<0.05);三仙中剂量组0.881mm较尼尔雌醇组0.737mm也显著升高(P<0.05)。E2、孕酮及子宫湿重:OVX组显著低于Sham组(P<0.001),尼尔雌醇组显示一定的升高趋势,三仙各组与OVX相比差异无显著性意义。体质量:OVX组400g较Sham组347g显著上升(P均<0.05);尼尔雌醇组较OVX组及三仙各组显著减轻(P<0.001);三仙各组与OVX相比无明显变化。结论尼尔雌醇对去卵巢大鼠骨密度和骨结构的改善无明显作用;三仙汤能够改善去卵巢大鼠骨生物力学性能,对OVX大鼠雌激素水平无明显影响。
Objective To investigate the effects of Sanxian decoction on bone mineral density (BMD), bone biomechanics, and serum estradiol (E2) and progesterone (P) levels in ovariectomized rats. Methods Sixty-eight 3-month-old female Sprague-Dawley rats were randomly divided into 7 groups: blank group, sham group (Sham), model group (OVX), nilestriol group, and three doses of Sanxian decoction. group. The BMD, maximum load (E), damage deflection (R), E2, P, body mass, and uterine wet weight were measured 3 months after the administration. Results Bone mineral density: OVX group 0.270g/cm2, significantly lower than Sham group 0.291g/cm2 (P<0.05), the middle dose of Sanxian had an upward trend compared with OVX. Maximum load: There was a certain increase in the 117kg medium dose of Sanxian and 114kg of the high dose group compared with 104kg of the OVX group. Destructive Deflection: The OVX group had a significantly lower trend than the Sham group; the three small, medium, and large dose groups of the Sanxian group were significantly higher than the OVX group (P<0.05); the 0.881mm group of the Sanxian medium-dose group was 0.737 compared with the Nilestriol group. Mm also increased significantly (P<0.05). E2, progesterone and uterine wet weight: The OVX group was significantly lower than the Sham group (P<0.001), and the nilestriol group showed a certain increase tendency. There was no significant difference between the three groups of the sham group and OVX. Body weight: 400g in OVX group was significantly higher than 347g in Sham group (P<0.05); Nilestriol group was significantly less than OVX group and Sanxian group (P<0.001); Sanxian group had no significant difference compared with OVX group. Variety. Conclusion Nilestriol has no significant effect on bone mineral density and bone structure in ovariectomized rats. Sanxian Decoction can improve the biomechanical properties of ovariectomized rats and has no significant effect on the level of estrogen in OVX rats.
