目的探讨ghrelin对TNF-α诱导的HBMEC人脑微血管内皮细胞PAI-1 mRNA的影响及意义。方法培养HBMEC人脑微血管内皮细胞,加入不同浓度的TNF-α(0.1ng/ml,1ng/ml,10ng/ml),采用逆转录-聚合酶链反应测定(reverse transcription-polymerase chain reaction,RT-PCR)检测PAI-1 mRNA的水平;ghrelin单独作用(1ng/ml,10ng/ml)或预处理1h后,加入TNF-α(1ng/ml)检测相应指标的变化。结果 TNF-α(0.1ng/ml,1ng/ml,10ng/ml)浓度依赖地增高人脑微血管内皮细胞PAI-1 mRNA的表达;而给与ghrelin单独作用(1ng/ml,10ng/ml)或预处理(10ng/ml)的人脑微血管内皮细胞PAI-1 mRNA的表达减低。结论 Ghrelin在HBMEC人脑微血管内皮细胞可抑制TNF-α诱导的PAI-1 mRNA表达,证实ghrelin在脑血管疾病的治疗中可能起重要作用。
Objective To investigate the effect and significance of ghrelin on PAI-1 mRNA expression in HBMEC human brain microvascular endothelial cells induced by TNF-α. Methods Human brain microvascular endothelial cells (HBMEC) were cultured with different concentration of TNF-α (0.1ng / ml, 1ng / ml, 10ng / ml) and reverse transcription-polymerase chain reaction (RT- PCR was used to detect the level of PAI-1 mRNA; TNF-α (1ng / ml) was added to detect the change of corresponding index after ghrelin alone or 1ng / ml or 1h pretreatment. Results TNF-α (0.1ng / ml, 1ng / ml, 10ng / ml) increased PAI-1 mRNA expression in human brain microvascular endothelial cells in a concentration- Pretreatment (10ng / ml) of human brain microvascular endothelial cells PAI-1 mRNA expression decreased. Conclusions Ghrelin inhibits TNF-α-induced PAI-1 mRNA expression in human brain microvascular endothelial cells (HBMECs), confirming that ghrelin may play an important role in the treatment of cerebrovascular diseases.
