测定5-氟脲嘧啶(5-Fu)血药浓度的方法有生物法、同位素标记法、分光光度法、气相法及高效液相法。本文采用氟离子选择电极对5-Fu 的血浓度作了初步测定,测定灵敏度为0.19μgF/ml,即相当于5-Fu 1.3μg/ml,血浓度的线性范围为10~90μg/ml血浆。1.直接燃烧法:在无灰滤纸上滴加400μl家兔血浆,吹干,用氧瓶燃烧后吸收于加有50μl3%H_2O_2的10ml重蒸水(或 pH 为5~6的醋酸缓冲液)中,用氟电极及带有硝酸钾外盐桥的饱和甘汞电极测定电位。2.提取燃烧法:取家兔血浆1 ml,加200μlpH
