目的观察抵当芪桂汤对STZ致糖尿病大鼠晶状体、晶状体上皮细胞以及HSP70水平的影响。方法随机抽取10只SD大鼠为空白对照组,其余大鼠一次性腹腔注射链脲佐菌素(STZ)60 mg/kg,复制糖尿病大鼠模型。选取成模动物55只随机分为模型组、达美康组、抵当芪桂汤组、抵当芪桂汤大剂量组、抵当芪桂汤加达美康组,分别给予相应干预。于实验2、4、6、8 W在裂隙灯下检查晶体混浊情况;8 W后显微镜下察晶状体上皮细胞形态变化,并检测晶状体上皮细胞(LEC)中的HSP70表达情况。结果空白对照组大鼠晶状体始终保持透明完整,模型组大鼠晶状体逐渐以致完全混浊,其余治疗组大鼠晶状体混浊程度由重到轻依次为抵当芪桂汤大剂量组、抵当芪桂汤组、达美康组、抵当芪桂汤加达美康组;HE染色晶状体上皮细胞形态亦发生不同改变;以空白对照组大鼠作为标准,模型组晶状体LEC中HSP70表达逐渐增多,治疗组抵当芪桂汤大剂量组HSP70的阳性表达率高于达美康组、抵当芪桂汤组、抵当芪桂汤加达美康组HSP70的阳性表达率,差异具有显著性(P<0.05)。结论抵当芪桂汤通过降低血糖、减少糖代谢产物在晶状体内聚集、降低晶状体内渗透压从而有效保护实验性糖尿病大鼠晶状体损伤,抑制糖尿病大鼠白内障的发生和发展,对早期糖尿病大鼠白内障具有一定的防治作用。
Objective To observe the effect of Daoguangji Decoction on lens, lens epithelial cells and HSP70 in STZ-induced diabetic rats. Methods Ten SD rats were randomly selected as the blank control group. The other rats were injected intraperitoneally with streptozotocin (STZ) 60 mg / kg to induce diabetic rats. Results In the control group, the lens of the rats in the control group remained clear and complete. The lens in the model group gradually became completely turbid. The degree of lens opacity in the rest of the rats in the control group changed from heavy to light to high dose of Qidang Decoction, The control group rats were used as the standard, and the expression of HSP70 in the LEC of the model group was gradually increased. The treatment group arrived at Qi Gui Tang The positive expression rate of HSP70 in high dose group was higher than that in DaMei Kang group, Dundang Qi Gui Tang group and Dundang Qi Gui Tonga Meikang group, the difference was significant (P <0.05). Conclusion Daoguang Decoction can effectively prevent the lens damage in diabetic rats and inhibit the occurrence and development of cataract in diabetic rats by decreasing blood glucose, reducing the accumulation of glucose metabolites in the lens, reducing intraocular lens osmotic pressure, Has a role in prevention and treatment.
