目的探讨前列腺癌(PCa)患者手术前后外周血CD4+CD25+调节性T细胞(Tregs)及其特异标志物FOXP3mRNA的表达比例的变化及临床意义。方法应用流式细胞术检测50例前列腺癌及50例正常对照组外周血单个核细胞(PBMC)中Tregs占CD4+T细胞的比例,应用RT-PCR技术检测人外周血PBMC FOXP3 mRNA的表达。结果随着Gleason分级升高,CD4+CD25+Tregs/CD4+T比值和FOXP3 mRNA表达量均有升高趋势;PCa患者术前CD4+CD25+Treg细胞占CD4+T细胞的比例和FOXP3 mRNA表达水平高于正常对照组(均P<0.05),术后水平与正常对照组差异无统计学意义;PCa组Tregs表达率与FOXP3 mRNA呈正相关(r=0.623,P<0.01)。结论 Tregs及其特异标志物FOXP3具有维持自身免疫稳定和抑制肿瘤免疫作用,可能参与前列腺癌的发生。
Objective To investigate the changes of the proportion of CD4 + CD25 + regulatory T cells (Tregs) and its specific marker FOXP3 mRNA in peripheral blood of patients with prostate cancer (PCa) and their clinical significance. Methods Flow cytometry was used to detect the percentage of Tregs in CD4 + T cells in peripheral blood mononuclear cells (PBMCs) from 50 cases of prostate cancer and 50 cases of normal controls. The expression of FOXP3 mRNA in PBMC of human was detected by RT-PCR. Results As the Gleason score increased, the ratio of CD4 + CD25 + Tregs / CD4 + T and FOXP3 mRNA increased. The percentage of preoperative CD4 + CD25 + Treg cells in CD4 + T cells and FOXP3 mRNA expression The level of Tregs in PCa group was positively correlated with FOXP3 mRNA (r = 0.623, P <0.01). The level of Tregs in PCa group was significantly higher than that in normal control group (all P <0.05). Conclusion Tregs and its specific marker FOXP3 may play an important role in maintaining autoimmune stability and inhibiting tumor immunity, and may be involved in the pathogenesis of prostate cancer.
