目的研究丹参冻干粉中原儿茶醛在孕羊体内的药代动力学过程。方法在清醒无应激条件下,母羊静脉给予不同剂量(5、10、20 mg/kg)丹参冻干粉,采用高效液相色谱内标法测定母羊静脉给予丹参冻干粉后不同时间(5、30、60、90、120、240 min)血浆中原儿茶醛的浓度,用Bapp6.0药动学软件计算其药代动力学参数。结果母羊静脉给予不同剂量丹参冻干粉后5、30、60、90 min均可测得母羊血浆中原儿茶醛浓度,且与丹参给药剂量成一定的线性关系。原儿茶醛的血浆半衰期为60 min左右,它在母羊体内吸收很快,达峰时间为5 min,而后以较快速度消除。结论研究丹参在母羊体内的药代动力学过程可为临床应用丹参防治妊娠期疾患时制定合理的给药方案提供依据。
Aim To study the pharmacokinetics of Salvia miltiorrhiza in pregnant sheep by freeze-dried powder of Radix Salviae Miltiorrhizae. Methods Freezing-dried Salvia miltiorrhiza powder (5, 10, 20 mg / kg) was intravenously administered to ewes under awake and no-stress condition. The eosinophils (5,30,60,90,120,240 min) plasma protoporphyrin concentration, with Bapp6.0 pharmacokinetic software to calculate the pharmacokinetic parameters. Results Echinacea concentration in ewe plasma could be measured at 5, 30, 60, and 90 min after intravenous administration of different doses of Danshen freeze-dried powder, and had a certain linear relationship with the dosage of Salvia miltiorrhiza. Protocatechualdehyde plasma half-life of 60 min or so, it quickly absorbed in the ewes, the peak time of 5 min, and then quickly eliminated. Conclusion The study of the pharmacokinetics of Salvia miltiorrhiza in ewes can provide a basis for the development of a reasonable dosing regimen for Salvia miltiorrhiza in the prevention and treatment of pregnancy disorders.
