目的:评价两种型号,三个浓度的内毒素经支气管滴注8h后所致急性肺损伤的大鼠模型。方法:Wistar雄性大鼠随机分为7组,麻醉后,经支气管分别滴注型号为L2630与L2880,浓度为1mg/kg,2mg/kg及3mg/kg的0111:B4与055:B5内毒素,建立大鼠急性肺损伤模型。造模8h后测定呼吸功能、血气分析、肺湿/干重(W/D)比值等。结果:型号为L2630与L2880,浓度为1mg/kg,2mg/kg及3mg/kg的内毒素经支气管滴注均可诱导大鼠急性肺损伤。结论:型号L2630,浓度为2mg/kg的内毒素支气管滴注8h所造成的急性肺损伤模型病理损伤和动物功能学指标最符合急性肺损伤模型的实验要求。
OBJECTIVE: To evaluate the rat model of acute lung injury induced by endotoxin of three models and three concentrations after bronchial instillation for 8h. Methods: Wistar male rats were randomly divided into 7 groups. After anesthesia, the rats were inoculated with endotoxin of 0111: B4 and 055: B5 with the concentrations of 1mg / kg, 2mg / kg and 3mg / Acute lung injury model in rats was established. Respiratory function, blood gas analysis, lung wet / dry weight (W / D) ratio and so on were measured 8 h after modeling. Results: Acute lung injury induced by endotoxin of L2630 and L2880 at concentrations of 1 mg / kg, 2 mg / kg and 3 mg / kg was induced by bronchial instillation. CONCLUSION: The pathological injury and animal functional indexes of acute lung injury model caused by L2630 and 8 mg / kg endotoxin bronchial instillation at 2mg / kg are most in line with the experimental requirements of acute lung injury model.
