目的:测定胃癌AGS细胞内pH(intracellular pH,pHi),探讨细胞外酸性环境对AGS细胞增殖和凋亡的影响。方法:采用离子成像分析系统测定AGS细胞的pHi,MTT及流式细胞术分别检测培养环境pH值为6.0时AGS细胞的活力、细胞周期和凋亡。结果:AGS细胞内pH明显碱化,在pH值为6.0的酸性环境培养时,细胞活力在12小时内明显抑制,细胞停滞在S和G2/M期,细胞凋亡不明显。结论:胃癌细胞AGS明显碱化,pH值为6.0的酸性环境使细胞数量减少和活力下降,其机制是使细胞出现增殖抑制而与凋亡无关。
OBJECTIVE: To determine the intracellular pH (pHi) of AGS in gastric cancer and investigate the effect of extracellular acidic environment on the proliferation and apoptosis of AGS cells. Methods: AGS cells were assayed by ion imaging analysis system. PHi, MTT and flow cytometry were used to detect AGS cell viability, cell cycle and apoptosis in the culture environment at pH 6.0. Results: The AGS intracellular pH was significantly alkalized. When cultured in acidic pH 6.0, the viability of AGS cells was significantly inhibited in 12 hours, and the cells arrested in S and G2 / M phases. The apoptosis of AGS cells was not obvious. CONCLUSIONS: The AGS of gastric cancer cells is significantly alkalized. Acidic pH value of 6.0 decreases the number of cells and decreases the viability. The mechanism is that the proliferation of cells is inhibited but not related to apoptosis.
