目的:研究不同剂量碱性成纤维细胞生长因子(bFGF)对人胚成骨样细胞DNA、胶原蛋白和碱性磷酸酶合成的影响。方法:实验分为4组,每组6孔,每组分别加入浓度为0.1ng/ml,1ng/ml,10ng/ml,100ng/ml的bFGF,每组均设空白对照,在实验后第1,3,5,7天分别检测人胚成骨样细胞3H-TdR和3H-脯氨酸的掺入量,以及碱性磷酸酶合成量。结果:bFGF可明显促进人胚成骨样细胞的DNA合成,但却抑制其胶原蛋白和碱性磷酸酶的合成,bFGF对人胚成骨样细胞具有双重效应。在含量为0.1~10ng/ml,呈剂量依赖性,10ng/ml时bFGF的作用最强。结论:bFGF在骨形成中有一定作用,但仍需与其他细胞生长因子协同作用
OBJECTIVE: To study the effects of basic fibroblast growth factor (bFGF) on the DNA, collagen and alkaline phosphatase synthesis of human osteoblast-like cells. Methods: The experiment was divided into 4 groups with 6 wells in each group. The bFGF with the concentration of 0.1ng / ml, 1ng / ml, 10ng / ml and 100ng / ml were added in each group. Blank control was set in each group. 3H-TdR and 3H-proline incorporation, and alkaline phosphatase synthesis of human osteoblast-like cells were detected on days 1, 3, 5 and 7, respectively. Results: bFGF promoted the DNA synthesis of human osteoblast-like cells, but inhibited the synthesis of collagen and alkaline phosphatase. BFGF had a dual effect on human osteoblast-like cells. In the dosage of 0.1 ~ 10ng / ml, in a dose-dependent, 10ng / ml bFGF strongest role. Conclusion: bFGF plays a role in bone formation, but it still needs to cooperate with other cell growth factors
