目的了解PCR用于鼠疫陈旧脏器材料的检测效果。方法按鼠疫诊断标准WS279-2008附录C的操作说明进行实验。结果 2005年的57份细菌学检验阳性的豚鼠脏器材料中,PCR检测结果检出645 bp、249 bp和456 bp 3条条带的为7份,显示645 bp和249 bp 2条条带的有32份,显示645 bp和456 bp 2条条带的有1份;其中3条条带均显示的仅为7份,占12.3%,作为阴性对照的30份达乌尔黄鼠脏器材料均只显示645 bp 1条条带。结论 PCR用于鼠疫陈旧脏器组织的检验时,检出率较低,长期保存的脏器材料可能存在核酸片段的破坏和丢失。
Objective To understand the effect of PCR on the detection of old organs of plague.
