目的 探讨TGF β对胎兔颅骨成骨样细胞体外成骨作用及BMP蛋白表达的影响。 方法 采用ALP活性测定及骨结节计数方法观察TGF β对胎兔颅骨成骨样细胞体外的成骨作用 ,BMP McAb免疫组化染色及其定量分析观察TGF β对胎兔颅骨成骨样细胞BMP蛋白表达的影响。结果 TGF β( 3~ 7d)对ALP活性有促进作用 ,并呈一定的时效和量效关系。TGF β对骨结节形成低剂量时有促进作用 ,高剂量时有较强的抑制作用 ;TGF β对成骨细胞BMP蛋白表达表现为抑制作用 ;二者均有显著性差异。 结论 TGF β对胎兔颅骨成骨样细胞体外成骨及BMP蛋白表达表现为双相作用。
Objective To investigate the effect of TGFβ on the osteogenic effect of osteoblast-like cells in vitro and the BMP protein expression in vitro. Methods ALP activity assay and bone nodule counting method were used to observe the effect of TGF β on osteoblast-like cells in vitro. BMP McAb immunohistochemistry and quantitative analysis were performed to observe the effects of TGF β on osteoblast-like cells Effect of protein expression. Results TGF β (3 ~ 7d) promoted the activity of ALP, and showed certain time-effect and dose-response relationship. TGFβ promoted the formation of low-dose bone nodules, and inhibited the expression of osteoblast BMP protein at high dose. TGF-β also inhibited the expression of BMP protein in osteoblasts. Both of them had significant difference. Conclusion TGF β has a biphasic effect on osteoblast-like osteoblast-like cells in vitro and BMP protein expression.
