目的:研究敏咳清颗粒对人胚肺成纤维细胞(LFB)产生转化生长因子-β1(TGF-β1)及Ⅲ型胶原(COL-Ⅲ)的影响,探讨敏咳清颗粒治疗咳嗽变异性哮喘的作用机制。方法:将常规培养LFB分为空白组、模型组、对照组、敏咳清高剂量组、敏咳清低剂量组、养阴清肺糖浆高剂量组、养阴清肺糖浆低剂量组,加入肝素刺激后分别加入对应的含药血清,采用ELISA法检测各组细胞培养上清液中TGF-β1、COL-Ⅲ的水平。结果:①敏咳清颗粒能明显减少TGF-β1的产生,高剂量组的作用明显强于低剂量组,高、低剂量相比有显著性意义(P<0.01);②敏咳清颗粒能明显抑制肺成纤维细胞表达COL-Ⅲ,高、低剂量组之间相比无显著意义。结论:敏咳清颗粒能显著降低经肝素刺激增殖的LFB对TGF-β1、COL-Ⅲ的表达,提示抑制TGF-β1、COL-Ⅲ的表达,可能是其治疗咳嗽变异型哮喘(CVA)的作用机制之一。
Objective: To study the effect of Minkeqing granules on the production of transforming growth factor-β1 (TGF-β1) and type III collagen (COL-III) in human fetal lung fibroblasts (LFB), and to investigate the effect of Minkeqing granule on cough variant asthma The mechanism of action. Methods: Routine culture LFB was divided into blank group, model group, control group, Minkeqing high dose group, Minkeqing low dose group, Yangyin Qingfei syrup high dose group, Yangyin Qingfei syrup low dose group, and heparin was added. After stimulation, the corresponding drug-containing serum was added, and the levels of TGF-β1 and COL-III in the cell culture supernatant of each group were detected by ELISA. Results: 1 Minkeqing granules can significantly reduce the production of TGF-β1, the effect of high-dose group was significantly stronger than that of low-dose group, and there was significant difference between high and low doses (P<0.01); The expression of COL-III in lung fibroblasts was significantly inhibited. There was no significant difference between high and low dose groups. Conclusion: Minkeqing granule can significantly reduce the expression of TGF-β1 and COL-III induced by heparin-stimulated LFB, suggesting that it inhibits the expression of TGF-β1 and COL-III, which may be the treatment of cough variant asthma (CVA). One of the mechanisms of action.
