目的 探讨人肾癌组织EGFR mRNA的转录水平、细胞定位及与肾癌生长和发展的关系。 方法 采用Digoxin标记原位杂交法 ,检测 2 5例石蜡包埋存档的肾癌组织和 2 0例相应正常肾组织及配对 16例冰冻保存的肾癌组织和远离癌自身正常肾组织标本的EGFR mRNA转录水平和细胞定位。 结果 肾癌组织和正常肾组织的EGFR mRNA转录阳性率分别为 :石蜡包埋组 44 % (11/2 5 )和 15 % (3/ 2 0 ) ;冰冻保存组 5 0 % (8/ 16 )和 13% (2 / 16 )。两组肾癌组织EGFR mRNA转录阳性率均较正常对照肾组织显著增高 (P均 <0 .0 5 ) ;癌组织原位杂交阳性见于癌细胞 ,主要位于胞核 ,少数位于胞浆 ;而两组正常对照肾组织为弱阳性反应。 结论 人肾癌组织过度转录EGFR mRNA ,可能与肾癌的生长、发展有关。
Objective To investigate the transcriptional level of EGFR mRNA, the location of EGFR mRNA in human renal cell carcinoma and its relationship with the growth and development of renal cell carcinoma. Methods Digoxin-labeled in situ hybridization was used to detect the expression of EGFR mRNA in 25 cases of paraffin-embedded renal cell carcinoma and 20 cases of normal renal tissue and paired 16 cases of cryo-preserved renal cell carcinoma and its normal renal tissue specimens Transcriptional level and cellular localization. Results The positive rates of EGFR mRNA in renal cell carcinoma and normal renal tissues were 44% (11/2 5) and 15% (3/20) in the paraffin embedded group and 50% (8/16) And 13% (2/16) respectively. The positive rates of EGFR mRNA transcripts in both groups were significantly higher than those in normal controls (all P <0.05). In situ hybridization of cancer tissues was found in cancer cells, mainly located in nucleus and few in cytoplasm. Group normal control renal tissue is weak positive reaction. Conclusion Overexpression of EGFR mRNA in human renal cell carcinoma may be related to the growth and development of renal cell carcinoma.
