目的 探讨 2型糖尿病患者尿中基质金属蛋白酶组织抑制剂水平的变化及其与糖尿病肾损害的关系。方法 采用酶联免疫吸附法对 78例 2型糖尿病患者及 30例正常人尿中基质金属蛋白酶组织抑制剂水平进行检测。结果 糖尿病各组患者尿中基质金属蛋白酶组织抑制剂水平均明显高于对照组 (P <0 .0 1) ;糖尿病患者各组间尿基质金属蛋白酶组织抑制剂水平差异显著 ,随 2 4h尿白蛋白排泄量的增加而递增 ,尿中基质金属蛋白酶组织抑制剂水平与 2 4h尿白蛋白排泄量、尿N 乙酰 β 氨基葡萄糖苷酶 (NAG)含量呈显著正相关。结论 糖尿病患者尿中基层蛋白酶组织抑制剂水平随糖尿病肾病的严重程度而明显增高 ,可作为糖尿病肾病早期诊断的敏感指标。
Objective To investigate the changes of urinary tissue inhibitor of matrix metalloproteinase (TIMP-1) and its relationship with diabetic nephropathy in type 2 diabetic patients. Methods The urinary levels of tissue inhibitor of matrix metalloproteinase (TIMP-1) in 78 patients with type 2 diabetes mellitus and 30 normal controls were detected by enzyme-linked immunosorbent assay. Results The urinary levels of MMP-2 and TIMP-1 in diabetic patients were significantly higher than those in the control group (P <0.01). The levels of urinary tissue inhibitor of tissue inhibitor of metalloproteinase (TIMP-1) Protein excretion increased with increasing urinary MMP inhibitor levels of 24 h urinary albumin excretion, urinary N acetyl β-glucosaminidase (NAG) content was significantly positively correlated. Conclusions The level of urinary basal tissue protease inhibitor in patients with diabetes mellitus is significantly increased with the severity of diabetic nephropathy and may be used as a sensitive indicator of early diagnosis of diabetic nephropathy.
