应用单克隆抗体(Mab)和 Dot-ELISA 方法检测血清中卫氏并殖吸虫(Pw)种、期特异性抗原(Ag),以进行并殖吸虫病活动性感染的血清学诊断。结果所有痰卵阳性的患者和大部分痰卵阴性的可疑患者血清中检出成虫期特异性 Ag。部分两类患者中血清还检出囊蚴期特异性 Ag。其它蠕虫感染、肺结核患者和健康中国人血清,囊蚴和成虫 Ag 均为阴性。以吡喹酮治疗实验感染犬可激发血清中 Pw 成虫 Ag 明显而短暂的增加,但在2个月内逐渐消失。本研究结果表明,我们建立的以自制 Mab 为探针的并殖吸虫 Ag 检测方法对于 Pw 活动性感染的诊断具有高度的敏感性和特异性。
The serological diagnosis of Paragonimus westermani (Pw) and stage specific antigen (Ag) in sera of paragonimiasis patients were performed by Mab and Dot-ELISA. Results All the sputum-positive patients and the majority of sputum-egg-negative suspicious serum were detected in adult stage Ag. Serum in some of the two patients also detected metaplasia-specific Ag. Other worm infections, tuberculosis and healthy Chinese serum, metacercaria and adult Ag were negative. Praziquantel treatment of experimental dogs can stimulate serum Pw Ag significant and transient increase, but gradually disappear within 2 months. The results of this study indicate that the Ag assay for paragonimiasis using a homemade Mab probe is highly sensitive and specific for the diagnosis of active Pw infection.
