目的:观察具有活血化瘀、补益肝肾作用的中药怀牛膝对重度颅脑损伤大鼠神经细胞凋亡的影响,探讨怀牛膝治疗颅脑损伤的作用机理。方法:以自制改良的Feeney’s自由落体打击装置造重度颅脑损伤大鼠模型,给怀牛膝水煎剂灌胃治疗1周,采用TUNEL法原位检测损伤区神经细胞的凋亡,观察怀牛膝对颅脑损伤大鼠神经细胞凋亡的影响。结果:与模型组相比,高剂量牛膝组治疗的颅脑损伤大鼠TUNEL阳性细胞数显著增多,P<0.05;其他各组的差异无统计学意义。结论:活血化瘀、补益肝肾的中药怀牛膝促进颅脑损伤大鼠神经细胞的凋亡,可能是怀牛膝改善了大鼠的免疫功能,增强了机体对受损神经细胞的清除能力,故凋亡细胞数增加。
OBJECTIVE: To observe the effect of Achyranthes bidentata which has the function of promoting blood circulation and removing blood stasis and tonifying liver and kidney on the apoptosis of nerve cells in rats with severe craniocerebral injury, and to explore the mechanism of Achyranthes bidentata treating craniocerebral injury. Methods: A rat model of severe traumatic brain injury was established by a self-made Feeney’s free-fall strike device. Gavage injection of Achyranthes bidentata decoction was administered intragastrically for 1 week. TUNEL method was used to detect the apoptosis of nerve cells in injured area. Effect of Knee on Neurocyte Apoptosis in Craniocerebral Injury Rats. Results: Compared with the model group, the number of TUNEL-positive cells in high dose Achyranthes group was significantly increased (P <0.05). There was no significant difference in other groups. CONCLUSION: Achyranthes bidentata (TCM), a Chinese herbal medicine for promoting blood circulation and removing blood stasis and tonifying the liver and kidney, can promote the apoptosis of nerve cells in rats with craniocerebral injury. It may be that Achyranthes bidentata improves the immune function of rats and enhances the ability of the body to remove the damaged nerve cells , So the number of apoptotic cells increased.
