目的探究紫花苜蓿黄酮在体外抑制肿瘤细胞活性的研究。方法采用四氮甲基唑蓝(MTT)法测定紫花苜蓿黄酮在体外的抗肿瘤活性;流式细胞术检测紫花苜蓿黄酮对人胃癌细胞MGC-803周期和凋亡的影响;Western blotting方法检测相关细胞蛋白的表达情况。结果 MGC-803细胞经紫花苜蓿黄酮预处理后,细胞存活率显著降低(P<0.05)。流式细胞术检测发现紫花苜蓿黄酮能够诱导细胞凋亡;引起细胞周期紊乱,使细胞阻滞在G0/G1期。Western blotting实验结果表明它能够活化促凋蛋白Caspase-9,Caspase-3,RARP,抑制抑凋蛋白Bcl-2的表达;引起相关细胞周期蛋白Cyclin D1,CDK4,CDK6的表达量下调。结论紫花苜蓿黄酮能够通过诱导MGC-803细胞凋亡途径和调节正常周期活动来抑制MGC-803细胞增殖,并且这种抑制作用还存在着时间和剂量依赖效应。
Objective To investigate the inhibitory effect of alfalfa flavonoids on tumor cell activity in vitro. Methods MTT assay was used to determine the antitumor activity of alfalfa flavonoids in vitro. Flow cytometry was used to detect the effect of alfalfa flavonoids on the cell cycle and apoptosis of human gastric cancer cell line MGC-803. Western blotting was used to detect the correlation Cell protein expression. Results The cell viability of MGC-803 cells pretreated with alfalfa flavonoids was significantly decreased (P <0.05). Flow cytometry found that alfalfa flavonoids can induce apoptosis; cause cell cycle disorders, so that the cells in the G0 / G1 phase. The result of Western blotting showed that it could activate caspase-9, Caspase-3, RARP and inhibit the expression of Bcl-2, and down-regulate the expression of Cyclin D1, CDK4 and CDK6. Conclusion Alfalfa flavonoids can inhibit the proliferation of MGC-803 cells by inducing the apoptosis pathway of MGC-803 cells and regulating the normal cycle activity. And this inhibition has time-and dose-dependent effects.
